Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) is a highly infectious and economically devastating virus affecting swine worldwide that causes Porcine Reproductive and Respiratory Syndrome (PRRS). Also known as pig “blue ear disease”, it is characterized by severe respiratory disease in piglets and reproductive failure in sows, causing major production losses.

Description of PRRSV
• Viral Characteristics: PRRSV is a positive-stranded RNA virus in the genus Betaarterivirus (family Arteriviridae).
• Genotypes: There are two distinct types, Type 1 (European) and Type 2 (North American), which share only 50–70% nucleotide similarity.
• Pathogenesis: The virus replicates in alveolar macrophages, the immune cells of the lungs, and spreads throughout the body, causing chronic infection and suppressing the immune system.
• Economic Impact: It causes roughly 1 billion dollars in damages annually in the US alone.

Major Antigens
The PRRSV genome contains 11-12 Open Reading Frames (ORFs) that encode structural and non-structural proteins (NSPs).
• ORF5 (GP5): The major envelope glycoprotein (GP5) is key to virus attachment to host cells. It is highly variable, making it a primary target for neutralizing antibodies, though its high mutation rate allows it to evade detection.
• ORF7 (N Protein): The nucleocapsid protein (N) is the most abundant and highly immunogenic protein. It is commonly used in diagnostic testing, although it does not induce strong neutralizing antibodies.
• ORF6 (M Protein): A non-glycosylated membrane protein that forms a complex with GP5 to aid in viral assembly.
• GP2, GP3 & GP4: Glycoproteins involved in virus entry.

Vaccine Development Status
• Current Limitations: Traditional vaccines, such as Modified Live Virus (MLV) and inactivated vaccines, provide incomplete protection against heterologous (different) field strains.
• Development Directions:
Subunit Vaccines: Research focuses on using subunits like GP5, M, and GP2, GP3, & GP4 glycoproteins to boost immunity.
