We offer 3 innovative vaccine adjuvants (CAS-1, CAS-2, & CAS-5) developed & produced by Clover Biopharmaceuticals for use in R&D.

Vaccine Adjuvants, also known as immunomodulators or immunoenhancers, can augment the immune response to an antigen. A vaccine often consists of an antigen and an adjuvant which acts as an “alarm bell” to:
1. Attract Immune Cells: It draws crucial immune cells (like dendritic cells and macrophages) to the injection site.
2. Enhance Antigen Presentation: It helps these immune cells better "eat" the antigen, process it, and present it to other key players, like the T-cells and B-cells.
3. Promote Long-Lasting Memory: This stronger initial response leads to the creation of more memory B-cells and T-cells, which are responsible for long-term immunity. This means the protection from the vaccine lasts longer. As an immune booster, an adjuvant may also help reduce an antigen dosage and the number of vaccinations needed.

CAS-1

Key Ingredients: Squalene, Tocopherol, Polysorbate 80
Introduction:
CAS-1 is an AS03-like oil-in-water adjuvant system developed by Clover, similar in formulation to the GSK AS03® adjuvant system**. It contains active ingredients including squalene and tocopherol, which can effectively mobilize and activate immune cells like APCs (antigen-presenting cells), thereby significantly enhancing vaccine efficacy and efficiency. A similar adjuvant was utilized against the 2009 global H1N1 influenza and COVID-19 pandemics.

Clover has conducted preclinical evaluations of multiple recombinant vaccine candidates containing the CAS-1 adjuvant, including GLP Tox studies for a S-Trimer broad-spectrum COVID-19 vaccine candidate. The COVID-19 vaccine candidate containing CAS-1 has also completed Phase I human clinical trials in humans in South Africa and demonstrated good safety and excellent immunogenicity.

CAS-1 has been selected to be part of the CEPI adjuvant library, the world's first library of vaccine-enhancing adjuvants.

CAS-2
Key Ingredients: Squalene, Sorbitan trioleate (Span 85), Polysorbate 80
Introduction:
CAS-2 is an MF59-like oil-in-water adjuvant system developed by Clover, similar in formulation to the Novartis MF59® adjuvant system**. It contains active ingredients including squalene and saponin 85. It can activate macrophages at the injection site to produce cytokines and chemokines, resulting in enhancement in Th1-type biased cellular immune response.

Key Ingredients: QS-21, MPL, DOPC, Cholesterol
Introduction:
CAS-5 is an AS01b-like innovative lipid-based adjuvant system developed by Clover, similar in formulation to the AS01b® adjuvant system** used in the GSK Shingrix vaccine. It contains active ingredients such as QS-21 and MPL, which can activate Toll-like receptor 4 (TLR4), induce the release of pro-inflammatory cytokines, and elicit a Th1-type cellular immune response.
